Title: Physical-chemical, biological, analytical method validation and development of semi-solid dosage form from SSchinus terebinthifolius RADDI extracts

Abstract: Schinus terebinthifolius Raddi is a typical tree of northeastern caatingas, find from Ceara to Rio Grande do Sul, besides Paraguay and Argentina. In the folk medicine, its barks was used as astringent, tonic and stimulant and also antimicrobial activity and regenerating tissue. Due to large utilization of this plant, the aim of this study was to realize the quality control, to evaluate the antimicrobial activity and antioxidant capacity of the crude extract of S. terebinthifolius and to produce a medicinal form containing the crude extract and validate the analytical methodology. The quality control of barks of the S. terebinthifolius was carried out based on pharmacopoeia assays: loss of drying (PD), determination of extractive (TE), granulometric analysis (GA) and total tannins content (TT). The results found for the barks were: PD = 11.8 ±0.4 (DPR%=3.5), TE = 17.59 ± 0.52 (DPR% = 2.95), mean diameter of 0.36 mm e TT = 11.14 ± 0.11 (DPR% = 1.02). Besides, the crude extract was evaluated in tannins and polyphenolics content, showing values of 32.0 ± 0.,26 (DPR% = 0.82) and 34.98± 0.29 (DPR% = 0.82), respectively. The analytical methodology of crude extract and and medicinal form produce were realized by spectrosphotometry UV-VIS and statistically validated . The antioxidant capacity of the crude extract was: 9.50±0.43 (DPR%=4.55), and it was statistically comparable with ascorbic acid 7.48±1.24 (CV%=16.6). The antimicrobial activity of the crude extract and the aqueous and ethyl-acetate fractions were testing against the yeast Candida albicans and C. parapsilosis, and the bacteria Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis and Staphylococcus aureus. The crude extract and fractions showed antimicrobial activity against the yeast. The ethyl-acetate fraction showed best antimicrobial activity against the bacteria. For the preliminary study of the stability, samples it had been tested in the times 0 and 24 hours, 7, 14, 21 and 30 days, when a decrease observed in polyphenolics content between the first one and the last test.
