Title: Seed extracts of Paullinia cupana: phytochemical evaluation, antioxidant CAPACITY and microbiological Preliminary study on the prevention of formation of dental plaque. 

Abstract: Paullinia cupana H. B. K. var. sorbilis (Mart.) Ducke, popularly known as “guaraná”, it is a Brazilian native plant. Its seeds are mainly used as stimulant, and they have a high concentration of methylxanthines and tannins. From the crude extract (EBPC) of the “guaraná” seeds were obtained two semipurified extracts FAQ and EPA (Required Patent PI#0.006.638-9). EPA was chromatographed by column chromatography (Sephadex® LH-20) and 24 subfractions were obtained. EPA also was chromatographed by multilayer countercurrent chromatography and 7 subfractions were obtained. The following compounds were isolated and identified: catechin, epicatechin, ent-epicatechin, procyanidins B1, B2, B3 and B4, procyanidin A2 and procyanidin C1. The antioxidant capacity in vitro of the aqueous, crude and semipurifieds extracts were evaluated being used the techniques of the phosphomolybdenum and DPPH•. All of the extracts differed significantly of the standard (vitamin C) in the evaluation of the phosphomolybdenum compound and DPPH• assay. The values obtained in DPPH• were IC50 (μg.ml-1): EBPC = 6,67, EPA = 5,23, FAQ = 14,97 and EA = 9,59. It was accomplished a microbiologic preliminary study in the prevention of the dental plaque formation by the test of the adherence to a glass surface face to Streptococcus mutans and for the assay of reduction of acids production. It was observed that EBPC and FAQ were the ones that presented better results, demonstrating that the extracts of Paullinia cupana can be used in the prevention of the dental bacterial plaque formation.
