Title: Healing activity of the crude extract of Poincianella pluviosa incorporated in hydrogels.

Abstract: Poincianella pluviosa has already been described as a healing of skin wounds incorporated in gel. In an attempt to prolong the contact of the drug with the wound, it was proposed in this study to evaluate wound healing using polymer films and carrier systems such as cyclodextrins. The work was divided in two parts: 1) Polymeric film containing crude extract of P. pluviosa (CE): characterization, healing and antimicrobial activity; 2) CE complexed with cyclodextrin (HPβCD): complex characterization and gel healing activity. The films were evaluated for their physical and mechanical properties and complexation of CE with HPβCD was confirmed by spectroscopic tests. The antimicrobial activity was determined with strains of Staphylococcus aureus. For the in vivo experiments, wounds were made on the back of rats and treated daily 4, 7, 10 and 14 days with the film or with the gel, according with the groups. In the end of each period, were made a skin permeation analysis and histological analysis for reepithelization and cell proliferation and for gel treatment the collagen was also analyzed. The statistical data were obtained by GraphPad Prism using the t test and Mann-Whitney. It was demonstrated that the presence of CE in the films increased the capacity of absorb water and decreased his resistance and permeability. The CE of the film permeated the skin, reaching the dermis, being able to influence the reepithelization at 7 and 14 days and stimulate the cell proliferation at 10 and 14 days of treatment, besides showing antimicrobial activity against strains of S. aureus. The complexation of CE with HPβCD was confirmed, suggesting its interaction with more nonpolar parts of the CE. In the CE-HPβCD gel treatment, there was permeation, reaching the dermis, but in the histological analysis, no benefits were demonstrated compared to the negative control. Films with CE of P. pluviosa can be an alternative for wound healing, protecting and repairing the tissue and giving comfort to the patient.
