Title: Ethnopharmacological assessment and toxicology preclinical in vivo of the crude extract from rhizomes of Limonium brasiliense.
Abstract: The herbal therapy has been the choice of many women to relieve menstrual disorders. Limonium brasiliense is among the plants of Brazilian biodiversity popularly used for the treatment of premenstrual syndrome and as a regulator of menstrual periods. Studies suggest the popular use of its roots however recent botanical descriptions confirm its as rhizomes. Considering the approach ethnopharmacological of this species, this study aimed to perform morphological and anatomical study of rhizomes of L. brasiliense, and to evaluate in vivo the security of the use and hormonal activity of its crude extract (CE). For the morphological and anatomical study histological sections of plant tissue were observed under light microscopy and scanning electron microscopy. The plant material was characterized as rhizome with the presence of internodes from buds in young tissues, presence of multilayered periderm, parenchyma rays, presence of aerenchyma, vascular bundles arranged radially and sclereids groups scattered throughout the cortex and pith. The assessment of acute toxicity was conducted in male and female Swiss mice, which determined a LD50 of 4.8 g/kg. The evaluation of the chronic toxicity showed that CE, even above the considered therapeutic doses, remains secured to use. The evaluation of mutagenicity with the micronucleus test showed mutagenic potential in doses above 2.0 g/kg. The evaluation of the hormonal activity was carried out in immature female rats. The results show delayed age to opening the vaginal canal in animals treated with CE at doses 12.5, 25, 50, 100 or 200 mg/kg when compared to PC group. The obtained results: the increase of the relative uterus weight, progesterone and estrogen dosage analyzed with the estrous cycle phase, predominance of the diestrus phase during treatment, as well as histological examination of the uterus, ovaries, and the vagina organs, the data with CE at doses 25, 50 or 100 mg/kg demonstrated a tendency of the progesterone action by a possible interference of estrogen action. The results determined as rhizome the organ of the plant used in folk medicine and demonstrate the crude extract of L. brasiliense is safe for using in the animal species and the tested doses, and indicated to an effect on the female reproductive system.
