Title: Evaluation of in vitro antimicrobial potential of Trichilia catigua and chemical characterization of aqueous oligomeric fraction
Abstract: Trichilia catigua A. Juss. (Meliaceae) is a tree known as "Catuaba" or "Catiguá" and is widely distributed in Brazil and commonly used in folk medicine. Studies with the barks of Trichilia catigua suggest that this plant has antimicrobial, antinociceptive, aphrodisiac, antioxidant, anti-depressant activities and preventive action against brain damage. This study aimed to separation and characterization of aqueous semipurified fraction obtained from the barks of Trichilia catigua by nuclear magnetic resonance (NMR) 13C, column chromatography and capillary electrophoresis which was also carried out the methodological development. In addition, we performed the microbiological screening against bacteria and fungi, being them Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Enterococcus faecium and Enterococcus faecium vancomycin-resistant (EFVR), Helicobacter pylori, Candida albicans, Candida parapsilosis, Candida tropicalis, Trichophyton mentagrophytes,Trichophyton rubrum and Microsporum gypseym. Microbiological analyzes were performed by determining the minimum inhibitory concentration (MIC), and H. pylori was also conducted turbidimetry and evaluation of bactericidal activity. It was first produced the crude extract (CE) of catuaba, which was subsequently partitioned giving the ethyl acetate fraction (EAF) and aqueous fraction (AQF). The chromatographic methods developed to AQF demonstrated efficacy in the characterization of oligomeric proanthocyanidin of T. catigua, and the 13C NMR spectrum suggested the presence of proanthocyanidins and cinchonains type I and II as oligo or polymeric compounds. Regarding the antimicrobial activity, the extracts showed significant results especially for E. faecium and EFVR with MIC from 312,5 to 156,5 µg/ml. An overall assessment, the EAF has shown better results, especially for Gram-positive strains and fungi. In determining turbidity, the AQF has shown considerable promise against H. pylori. The results suggest that T. catigua extracts have potential application as an adjunct to treatment and promising to develop new antimicrobial drugs.
