Title: Nitro-compound identification and dihydro flavonoids, antibacterial, antifungal and antiviral compounds isolated from freeze-dried extracts of Heteropteris aphrodisiaca roots O. Mach., Malpighiaceae, Nó-de-cachorro.
Abstract: The vegetable Heteropteris aphrodisiaca O. Mach., belonging to the family Malphighiaceae popularly is known as "nó-de-cachorro", a bush from 1 to 1,5 m of height that can be found in the savannahs of the states of Mato Grosso and Goiás whose roots are irregular, with wrinkles and articulations and it has been used thoroughly by the popular medicine as aphrodisiac, stimulant, reviver, antioxidant, anticholesterolemic and in the nervous weaknesses. Of the extract BST-0298, it was partitioned with ethyl acetate, and was cromatographed by column was obtained after crystallization three compounds di-hidroflavonols: astilbin [3-O-α-L-ramnosil-trans-(2R,3R)-taxifolin]; neoastilbin [3-O-α-L-ramnosil-trans-(2S,3S)-taxifolin]; isoastilbin [3-O-α-L-ramnosil-cis-(2R,3S)-taxifolin]. Of the extract BST-0402, after column chromatographic and crystallization, it was isolated a nitrocompound: 2,3,4,6-tetra-O-(3-nitropropanoyl)-β-D-glucopiranosyl. The structures were elucidated with help the data of IR, MS, ¹H NMR, ¹³C NMR, DEPT, COSY, and HETCOR. The isolation of new nitrocompound is significant of the chemotaxonomic point of view. The crude extracts as well as the isolated compounds of Heteropteris aphrodisiaca were tested for antibacterial, for antifungic and antiviral activity. The nitrocompound lV presented a MIC against Staphylococcus aureus and Bacillus subtilis at the concentrations of 250 and 125 µg/ml and MBC resulted in 500 µg/ml, respectively. The antifungic activity resulted MIC at the concentration of 125 µg/ml against Candida albicans, C. parapslosis, C. krusei and C. tropicalis. MFC against the same organisms was shown at the concentration of 250 µg/ml. The nitrocompound lV revealed antiviral activity for the method TCID50 against Poliovírus Hep2 at the concentration of 50 mg/mL, presenting 69% of inhibition viral. The therapeutic antiviral activity by the method TCID50 at the concentrations of 12.5 and 25 mg/ml, being obtained significant result of 94 and 91% of viral, inhibition respectively. At the concentration of 50 mg/ml, the observed result was negative. The samples of crude extract and di-hidroflavonóis didn't present significant results in those analyses.
