Title: Quality evaluation, chemical study, histological and skin permeation of extracts from the bark of Guazuma ulmifolia LAM. VAR. tomentella, Sterculiaceae.

Abstract: Barks of “mutamba”, Guazuma ulmifolia Lam. var. tomentella, has been popularly used for the treatment of gastrintestinal illness, skin diseases and against hair fall. Quality control of the vegetable drug collected in two different seasons (winter and summer) was followed by pharmacopoeial assays. Phytochemical screening indicated the presence of taninns, flavonoids and saponins. The loss on drying obtained from the fresh winter drug (WD) was 56.59%, for the fresh summer drug (SD) this analyses was not done. Loss on drying obtained from the milled drug (winter and summer) and done after 2, 5, 12 months was below 13.5%. The best results for the solvents according to the determination of extractives was the acetone:water (1:1) for WD (16.56%) and acetone:water (7:3) for SD (10.97%). According to dry reside the best solvent was acetone:water (7:3) for the both drugs (0.1683 g and 0.1425 g). The flavonoids were obtained 0,0044% for WD and 0,0090% for SD. The mean diameter of granulomentric analyses assayed was calculated for WD (0.373 mm) and for SD (0.377 mm). There were isolated and identified nine substances from a semipurified extract: catechin, epicatechin, gallocatechin, epigallocatechin, 4´-O-methyl-epiafzelechin (new substance), epiafzelechin-(4β→8)-epicatechin, epicatechin-(4β→8)-catechin (PB1), epicatechin-(4β→8)-epicatechin (PB2), and epicatechin-(4β→8)-epigallocatechin. For the in vitro antioxidant capacity evaluation, two technics were made (phosphomolibdenum and DPPH) with crude and semipurified extracts. All extracts were statistically significant by phosphomolibdenum assay against C vitamin as standard. Among all the samples tested in the DPPH technic, the best results were obtained with the ethyl-acetate phase (IC50:  WD= 3,409 μg ml-1 e SD= 3,227 μg ml-1), both differed significantly to the standard (C vitamin). A capilar lotion was prepared to be used in a study about  hair growing activity in wistar rats. A morphological avaliation (microscopic) were realized through the morphometric and the epitelial celular proliferation analysis, but there were no evidence of the development of the hair follicle attributed, popularly, to “mutamba”. In the photoacoustic spectrometry was demonstrated the permeation of the pharmaceutical formulation.
