Title: Quality control and phytochemical evaluation of extract and fractions obtained from Paullinia cupana Kunth seeds and its activity against Helicobacter pylori
Abstract: Paullinia cupana Kunth, known as guarana, is a native Brazilian plant from the Amazon, and its seeds have wide use in the food and pharmaceutical market. Several biological activities have been described in the literature by using crude extracts and their semi-purified fractions. The objective of this study was to evaluate the quality of two different lots of the plant, chemically characterize the aqueous semipurified fraction (FAQ), and assess the activity of the crude extract and semipurified fractions against Helicobacter pylori. From the crude extract of P. cupanaa (EBPC) yielded the semipurified fractions in ethyl acetate (EPA) and water (FAQ). For quality control, of the two lots, pharmacopoeic analyses were made in plant drugs and analysis of chemical profile via HPLC-DAD was made in the EPA fraction. The chemical profile of the FAQ were performed by using 13C NMR and MS by direct infusion. The results of physico-chemical analyses for lots 1 and 2 were, respectively, 32.75% and 29.71% in extractives content; 1.87% and 1.42% of total ash; 8.84% and 9.94% loss on drying; 6.37% and 5.13% in the determination of methylxanthines, and 9.62% to 9.25% in total polyphenol content, and by HPLC-DAD showed similarity in the chemical profile of two lots. The solid 13C NMR spectrum showed characteristics of flavan-3-ol as signals corresponding to the two phenolic regions (rings A and B); bond of interflavonoids, these characteristics suggest the presence of condensed tannins (proanthocyanidins). The mass spectrum showed the presence of monomers, dimers, trimers and tetramers of procyanidins, and the presence of caffeic acid and derivatives. The biological evaluation showed that the extract and fractions do not have strong activity against Helicobacter pylori, but a bacteriostatic activity. The results demonstrate the quality and characteristics of the plant material used in the study and the presence of condensed tannins and caffeic acid in the FAQ guarana.
